Job: Molecular Cell Research Associate, Locus Biosciences, Morrisville, NC
Locus Biosciences is seeking a highly motivated and entrepreneurial applicant to join our research and development team advancing novel human gene editing technologies as a full-time Research Associate. This position is well suited for those that can accomplish defined work, as well as assist in approaching and solving complex scientific problems. This individual will be responsible for assisting in the design, generation, and characterization of novel human gene editing preclinical leads. The ideal candidate will be able to conduct general molecular and cellular biology research with supervisory oversight. This individual must be able to communicate effectively in formal and informal settings and be able to work well within a team. Candidates should have prior laboratory experience and be competent at basic molecular biology and tissue culture techniques. Candidates must be willing to navigate uncharted waters typical of a startup and possess a strong desire to embrace a high drive team culture. Seniority level may be negotiable depending on years of experience and technical competency.
Ideal candidates will possess a solid understanding and/or working experience with the following techniques:
Molecular biology – Mammalian expression vector design and engineering, testing and characterization of novel Type I CRISPR genetic engineering tools and vectors for working with a diverse range of human gene editing designs. Command of basic molecular biology techniques including DNA ligation, restriction enzyme digestion, gel electrophoresis, QPCR, RNA and DNA purification and quantification, cDNA synthesis and analysis, DNA sequence analysis.
Mammalian cell culture – Hands-on experience culturing human cell lines following best practices including; aseptic technique, passaging and preparing frozen vials, preparing sterile media, monitoring cell density and health, transfection protocols, cell harvest and processing RNA, DNA and protein for analysis. Expertise in transfection protocols and enrichment for edited cell populations by selection media or FACS is desirable.
Protein analysis – Experienced in basic biochemical assays for protein functional and structural characterization including; protein quantification, Western blotting, co-immunoprecipitation of multimeric effector complexes, cellular localization by immunofluorescence, protein-DNA binding assays, nuclease functional assays, functional characterization of fusion proteins such as epitope tags, NLS tags, and fluorescent reporters.
Laboratory upkeep – Candidate must have a solid understanding of laboratory maintenance, re-stocking, clean up, equipment cleaning and maintenance, and an overall willingness to pitch in to keep the lab running smoothly.
Candidate must possess:
A BS/ MS in molecular biology, genetic engineering or similar fields
At least 6 months full time experience working in a research laboratory setting
Familiarity with general laboratory techniques such as pipetting, PCR, RNA, DNA and protein manipulations, aseptic technique, common solution preparation
Working knowledge and experience with mammalian cell tissue culture techniques including passaging and maintaining cell lines and conducting transfection experiments
Ability to independently carry out assigned experiments and generate relevant data outputs for analysis and documentation in eNotebook
At least two years’ experience between academic or industry research
Ability to assist in the design of experiments and interpretation of results
Technical proficiency with designing and executing molecular biology experiments (PCR, transformation, Gibson cloning, RE cloning)
Hands-on mammalian cell line culturing and manipulation including CRISPR editing
Experience with computational tools analyzing DNA sequence (plasmid expression vector, genomic DNA, gene editing resulting from NHEJ and HDR) is highly desirable
