Job Description

StrideBio is a product-driven, fully integrated gene therapy company focused on creating and developing innovative genetic medicines with life-changing or curative potential for patients with devastating conditions, including monogenic rare disease and beyond.

We leverage our proprietary structure-inspired adeno-associated virus (AAV) vector engineering platform (STRIVE) to create unique and differentiated vectors that improve upon naturally occurring AAV serotypes to overcome current limitations of first-generation gene therapies. Combined with our genetic construct design expertise and in-house manufacturing capabilities, StrideBio is positioned to generate best-in-class genetic medicines that allow more patients to benefit with maximum efficiency, leading to improved treatment outcomes.

Our team is comprised of accomplished leaders and talented, passionate colleagues with a desire to translate innovative science into treatments. Together with our collaborators we are making rapid progress on our mission to bring novel medicines to patients who desperately need them. StrideBio's offices, labs and manufacturing facility are located in the heart of Research Triangle Park, NC, within 10 miles of downtown Durham and Raleigh-Durham International Airport. We seek a motivated molecular biologist with bioinformatics experience to join the Therapeutic Discovery group responsible for discovering lead recombinant adeno-associated virus (rAAV) vectors for preclinical development. You will be responsible for contributing to and optimizing rAAV expression cassette designs across multiple programs to enable the discovery and evaluation of novel rAAV therapies. You will also be expected to provide relevant bioinformatic support that will require broad collaboration across multiple functional groups (i.e., discovery, translational, process development, and QC).

Utilize deep understanding of RNA biology and regulatory mechanisms to inform cassette design and optimize transgene expression
Contribute to and execute codon-optimization strategies to enhance RNA stability and expression
Design/analyze/interpret RNAseq experiments for phenotype ID / correction, cell line screening / characterization
Analyze publicly available datasets for disease signature ID and target discovery
Analyze amplicon sequencing data to enable novel capsid evolution and discovery
Apply various NGS platforms to confirm packaged transgene sequences for Process Development and QC work
Verify and harmonize sequences of registered plasmids and cassette elements
Manage staff, including intern / contractors, for the execution of defined bioinformatics projects
Ensure accurate and up-to-date records of experimental activities via electronic documentation
Technical writing of procedures, protocols, and reports

Job Requirements

PhD in molecular biology, computational biology, or related field with 3 years of experience
Previous wet lab experience with RNAseq library preparation and molecular and cloning techniques (i.e. PCR, transfection, transduction)
Familiarity with human genomics, standard genomic databases (Ensembl and NCBI) and analysis tools
Experience with analyzing and interpreting large-scale genomic data, i.e. whole genome sequencing, amplicon sequencing, RNA-seq, MiSeq
Experience with one or more coding languages (i.e., Python)
Expertise in data analysis and visualization tools (i.e. R, Spotfire)
Strong written and verbal communications, including critical thinking skills
Demonstrated experience working independently, as well as effectively collaborating across functional groups
Demonstrated problem-solver who thrives on solving scientific challenges
Self-motivated and driven professional who can effectively perform in a rapidly changing, startup environment
